Understanding the Blood-Brain Barrier

The blood-brain barrier (BBB) is one of the most elegant systems in human physiology — a highly selective interface between the bloodstream and the central nervous system that lets nutrients through while blocking most toxins, pathogens, and even many medications.

What It's Made Of

The BBB is formed by endothelial cells lining brain capillaries, joined together by unusually tight junctions. Around them sit astrocytes (a type of glial cell) and pericytes, which together regulate what crosses. The result is a barrier far more selective than the vessels supplying, say, your liver.

What Crosses Easily

• Water (through channels)
• Oxygen and CO₂
• Small lipid-soluble molecules (including caffeine and alcohol)
• Glucose (via dedicated transporters)
• Certain amino acids (via active transport)

What Doesn't

• Large hydrophilic molecules
• Most bacteria and larger pathogens
• Many pharmaceuticals — one of the biggest challenges in neuroscience drug development

Why It Matters for Supplements

For a nutritional ingredient to have a cognitive effect, it has to either cross the BBB itself or influence something that does. L-Arginine, L-Citrulline, and Niacin all have well-characterised transport mechanisms into the brain. Beta-Alanine crosses more slowly but does reach nervous tissue, where it contributes to carnosine synthesis.

Aging and the BBB

The BBB becomes slightly more permeable with age — sometimes allowing molecules through that wouldn't have crossed at 30. This is one of the mechanisms behind age-related neuroinflammation.
